        • Transpupillary thermotherapy in the management of exudative age-related macular degeneration

          Objective lt;brgt;To evaluated the effect of transpupillary thermotherapy (TTT) on age-related macular degeneration (AMD). lt;brgt; lt;brgt;Methods lt;brgt;Sixty-two cases (62 eyes) of exudative AMD were managed with TTT. Before treatment, 58 cases underwent fundus fluorescein angiography(FFA),42 cases underwent simultaneous indocyanine green angiography (ICGA), and 56 cases underwent optic coherence tomography (OCT).TTT was delivered using a 810 nm diode laser with variable spot sizes 0.5-3.0 mm and power range 60-40 mW,60 seconds duration. Sixty-two cases were followed up for 1-10 months with 4.8 months average. lt;brgt; lt;brgt;Results lt;brgt;The visual acuities of last visit were compared with those before the treatment. The visual acuity was unchanged in 43 cases (69.3%), improved in 15 cases (24.2%), and declined in 4 cases (6.5%). OCT was re-done in 51 cases and compared with OCT images before TTT treatment. The height of macular edema was unchanged in 29 cases (56.9%), decreased in 18 cases (35.3%), and increased in 4 cases (7.8%). The amelioration of visual acuity was compatible with that of macular configuration in the majority of cases (74.5%). Only in 13 cases (25.5%) the amelioration of visual acuity lagged behind that of macular configuration. The re-treatment was performed in 18 cases (29.1%), probably due to insufficiency of laser power. No side-effect was found. lt;brgt; lt;brgt;Conclusion lt;brgt;TTT makes most of the cases of exudative AMD retaining or improving their visual acuity. The employment is secured.         • A MODIFIED OBSTRUCTING-PRESSING THERAPY FOR THE TREATMENT OF PATIE NTS WITH EXTERNAL INTESTINAL FISTULAS

          A modified abstructingpressing therapy done on 14 patients with high or low external intestinal fistulas following postoperative infection after abdominal operations was reported. The results showed that high or low external intestinal fistulas could be cured by the combined method of obstruction and compression. This therapy decreased the mortality rate, saved the patients from the risk period and shortened curing course of treatment. Finally the characteristics, the surgical procedure, precautions and advantages of the method were disscussed.

        • Alginate sodiumretinoic acid microspheres suppress the subretinal proliferation after laser coagulation

          Objective To observe the inhibiting effects of alginate sodiumretinoic acid(AGS-RA)microspheres release system on the laser coagulationinduced subretinal proliferation.Methods RA were dissolved by absolute alcohol,then mixed with 1.5% AGS and made into AGSRA microspheres by a microcapsule electrostatic generator. The parameter of laser injury include irradiation time (0.20 s),spot diameter (200 mu;m) and output power (420 mW).Thirty pigmented rabbits were randomly divided into 3 groups (laser injury,experimental and control group).After laser coagulation,AGSRA or blank microspheres were immediately injected into the vitrous of experimental and control rabbits respectively.The height,width and area of 6 retinal spots of laser coagulation at each timepoint were analyzed histopathologically with serial retinal sections at 1,2,3,4,and 6 weeks after laser coagulation.Results Histopathological examination showed that there were morphological and distribution changes of retinal cells in all layers, and localized fibroblasts proliferation in the retina after laser injury. The laserinduced responses in experimental group were much milder(P<0.01), while the laser injury group and control group have same width(P>0.05)and height/area of laser spots(P>0.05).Conclusion AGSRA release system can alleviate the subretinal proliferate after laser injury.

        • Research Progress on Roles of Survivin Gene in Tumor

          ObjectiveTo summarize the role of survivin gene in tumor. MethodsThe research status on biological characteristics the role of survivin gene in tumor for gene therapy and clinical application was retrospectively analyzed after related domestic and foreign literatures were reviewed. ResultsSurvivin gene was by far found to be the best powerful apoptosis inhibition gene, which played antiapoptosis role mainly through inhibiting directly the activity of caspase-3 and caspase-7 in the downstream of cascade reaction. Survivin gene promoted tumor cell proliferation and differentiation through speeding up tumor cells transition of G1→S phase and eluding the recognition of tumor cells to the apoptosis in G2/M phase. Survivin gene played important role in the intermediate links of vasiformation through angiogenic factor (VEGF, bFGF, Ang-1, and COX-2). ConclusionSurvivin gene may inhibit the apoptosis, promote the proliferation and differentiation of tumor cells and tumor angiogenesis, suggested that survivin gene has potential to act as a novel tumor marker and become an indicator of malignant tumor.

        • In Vivo Antitumor Activity of HSV tk/CD Combinative Gene Toward Human Cholangiocarcinoma Cells(QBC939)

          【Abstract】Objective To study the antitumor activity of HSVtk/CD combinative gene toward human cholangiocarcinoma in vivo. Methods Nude mouse models with transplanted subcutaneous cholangiocarcinoma were constructed and divided into 4 groups randomly, each group had 8 mice. Adenovirus solution free from suicide gene was injected in subcutaneous tumors of each mouse of control group. Adenovirus solution containing cytosine deaminase (CD), thymidine kinase (tk) and HSVtk/CD fusional gene were injected into single suicide gene either HSVtk or CD was transinfected into the tumor cells by injecting viras into subcutaneous tumor of mice of CD gene,tk gene and fused CD and tk gene group respectively. 24 hours after the injection, 5fluorocytosine (5FC) and ganciclovir (GCV) were injected introabdominally in each mouse. Growth of the tumors were monitored.Results Tumor growth of the genetransfection groups was suppressed in different degrees. Compared with the control group, the suppressing rates of the genetransfection groups were 41.2%, 55.7% and 70.0% respectively (P<0.05). Histological examination showed good tumor growth in the control group, and tumor necrosis in the other 3 groups, particularly obvious in the group transfected with pAd(HSVtk/CD).Conclusion Combinative gene system has a b antitumor effect on cholangiocarcinoma in vivo. But it’s not powerful enough to eliminate tumor thoroughly because of insufficient “Bystander effect”.

        • Change of BAG 3 Expression of Pancreatic cancer Tissues and Cell Lines after Chemotherapy

          【Abstract】Objective To analyze the function of BAG3 in antiapoptosis and chemotherapy resistance induction process of pancreatic cancer.Methods The expressions of BAG-3 in pancreatic cancerous tissues of patients with chemotherapy and those without chemotherapy before resection were determined by immunohistochemistry. The expression difference of BAG-3 protein 18 hours after cultured with chemotherapy drugs (concentration of drugs: 5-FU 50 μg/ml, MMC 0.5 μg/ml, EADM 1.5 μg/ml) of 3 pancreatic cancer cell lines (MIACaPa-2, PANC-1, SW1990) was measured through Western blotting method.Results The median positive rate of pancreatic cancer tissue from patients accepted chemotherapy before resection was higher than those not accepted chemotherapy, but there wasn’t significant difference. Eighteen hours after cultured with drugs, the level of BAG-3 of this three cell lines had significant increased compared with control group (P<0.05). Conclusion Chemotherapy induces elevation of BAG-3 expression of pancreatic cancer. The upregulate of BAG-3 may associate with the chemotherapy resistance induced by drugs.

        • Value of Chemosensitivity Assay In Vitro on Breast Cancer

          Objective To explore the value of chemosensitivity assay in vitro on breast cancer. Methods In vitro chemosensitivity of 6 species of chemotherapeutic agents applied to 38 cases of breast cancer patients were detected by tissue culture-end point staining-computer image analysis (TECIA). Results The sensitivity to chemotherapeutic agents commonly used in the breast cancer level from high to low was as follow: Doxorubicin (ADM), Paclitaxel (TAX), Vinorelbine (NVB), Cyclophosphamide (CTX), Cisplatin (DDP) and Fluorouracil (FU). Conclusion Drugs sensitivity experiment of cancer in vitro by TECIA has an important value to instruct clinical medication and individual chemotherapy for breast cancer.

        • Effects of Recombinant Adenovirus-Mediated Overexpression of hTIMP-1 on Proliferation of Human Liver Cancer Cell Line HepG2 in Vitro

          Objective  To explore the effects of overexpression of human tissue inhibitors of metalloproteinase-1 (hTIMP-1) on proliferation of human liver cancer cell line HepG2 in vitro. Methods A recombinant adenoviral vector containing full-length cDNA of hTIMP-1 was generated and transfected into HepG2. The viral titer was checked by measuring GFP, and the expression of hTIMP-1 in vitro was detected by the techniques of Western blot and semi-quantitative RT-PCR. The ultrastructure was observed by transmission electron microscope and the effects of overexpression of hTIMP-1 on proliferation of HepG2 in vitro was analyzed by MTT assay and growth curve. Results The resultant AdhTIMP-1 was successfully constructed and the expression of hTIMP-1 was detected by Western blot and RT-PCR. The growth and proliferation of HepG2, which had been transfected with AdhTIMP-1,  was significantly inhibited. Conclusion The proliferation of HepG2 was markedly inhibited by recombinant adenovirus-mediated overexpression of hTIMP-1, which may pave the way for further application in liver gene therapy.

        • Inhibitory effect of small interfering RNA targeting peroxisome-proliferator-activated receptor-γcoactivator-1αon retinal neovascularization in the mouse

          ObjectiveTo evaluate the inhibitory effect of small interfering RNA (siRNA) targeting peroxisome-proliferator-activated receptor-γcoactivator-1α(PGC-1α) on retinal neovascularization in the mouse. MethodsEighty seven-day-old C57BL/6J mice were divided into normal group, model blank group, model control group and PGC-1αsiRNA group, twenty mice in each group. Mice in the normal group were kept in normal room air. Mice in the model blank group, model control group and PGC-1αsiRNA group were induced for retinal neovascularization by hypoxia. Liposome with PGC-1αsiRNA (1 μl) and liposome with negative control siRNA (1 μl) were injected into the vitreous in the PGC-1αsiRNA group and model control group respectively when mice were moved out to room air from the cabin (Postnatal 12). No injection were performed in the model blank group. At postnatal 17, fluorescein angiography was used to assess the vascular pattern.The proliferative neovascular response was quantified by counting the nuclei of new vessels extending from the retina into the vitreous in cross-sections. PGC-1αand vascular endothelial growth factor (VEGF) level in retina were measured by real-time polymerase chain reaction (real-time PCR) and Western blot. Inhibition efficiency of PGC-1αsiRNA on PGC-1αand VEGF was calculated. ResultsMice in the normal group showed reticular distribution of retinal blood vessels. Central nonperfused retina, neovascular tufts and fluorescein leakage were seen in the model blank group and model control group. Neovascular tuft and fluorescein leakage were decreased in the PGC-1αsiRNA group compared to the model blank group and model control group. The neovascular nuclei were increased in the model blank group and model control group compared to the normal group (P < 0.05). The neovascular nuclei were decreased in the PGC-1αsiRNA group compared to the model blank group and model control group (P < 0.05). The expression of PGC-1αmRNA and protein in retina was increased significantly in the model blank group and model control group as compared with normal group, while decreased 54% and 53% respectively in the PGC-1αsiRNA group as compared with model blank group and model control group (P < 0.05). The expression of VEGF mRNA and protein in retina was increased significantly in the model blank group and model control group as compared with normal group, while decreased significantly in the PGC-1αsiRNA group (decreased 48% and 40% respectively) as compared with model blank group and model control group (P < 0.05). ConclusionsIntravitreal injection of PGC-1αsiRNA mediated by liposome can inhibit retinal neovascularization in the mouse effectively.

        • Research progress of circRNAs in thyroid papillary carcinoma

          ObjectiveTo summarize the role of circular RNA (circRNAs) in thyroid papillary carcinoma (PTC) and the emphasis of future research.MethodRelevant literatures in recent years about the biological function of circRNA and its role in PTC were reviewed.ResultscircRNAs had abnormal expression in PTC tissues. Besides, by working as miRNA sponges or interacting with RNA-binding proteins, circRNAs regulated the expression of proteins that were associated with cell proliferation, apoptosis, invasion, and metastasis, which could affect the biological characteristics of tumor cells.ConclusioncircRNAs are expected to be the biomarkers for early diagnosis of PTC or potential targets for PTC therapy and provid therapeutic bases to prevent PTC.
