化療對胰腺癌組織及細胞中BAG-3表達的影響_《中國普外基礎與臨床雜志》

【摘要】目的初步分析BAG3在胰腺癌抗凋亡及產生耐藥特性中的作用。方法通過免疫組化方法檢測術前化療與未化療患者手術切除的胰腺癌組織中BAG3的表達水平；通過蛋白質印跡方法檢測MIACaPa2、 PANC1及SW1990 3株細胞在化療藥物（5FU 50 μg/ml、 MMC 0.5 μg/ml及EADM 1.5 μg/ml）作用18 h后BAG3蛋白表達的差異。結果術前化療患者的胰腺癌組織中BAG3呈中強度陽性表達者的比率與術前未化療患者相比有升高趨勢，但差異無統計學意義（P gt;0.05）。化療藥物處理后18 h，3株胰腺癌細胞中BAG3的表達水平與未經化療藥物處理的對照組相比明顯升高（P＜0.05）。結論化療可使胰腺癌細胞中BAG3的表達升高，BAG3的上調可能與胰腺癌對化療產生耐藥相關。
