        • Expressions of Osteopontin and Related Factors in Breast Cancer Tissues and Its Clinical Significance

          Objective To detect the expressions of osteopontin (OPN), breast tumor kinase (Brk), and vascular endothelial growth factor (VEGF) in the breast cancer tissue, the adjacent (2cm) normal breast tissue, and the distal(>5cm) normal breast tissue, and analyze their clinical significances. Method The immunohistochemical method was used to detect the expressions of OPN, Brk, and VEGF in the breast cancer tissue, the adjacent (2cm) normal breast tissue, and the distal (>5cm) normal breast tissue from 40 cases of breast cancer. Results ① The expressions of OPN,Brk, and VEGF in the breast cancer tissue were significantly higher than those of the adjacent (2cm) normal breast tissue and the distal (>5cm) normal breast tissue (P<0.01), the expression of Brk in the adjacent (2cm) normal breast tissue was significantly higher than that of the distal (>5 cm) normal breast tissue (P<0.05). ② In the breast cancer tissue, the OPN and Brk protein expressions were not associated with age, tumor diameter, and histological grade (P>0.05),were associated with lymph node metastasis and TNM stage (P<0.05). The VEGF protein expression was not associated with age and tumor diameter (P>0.05), but was associated with histological grade, lymph node metastasis, and TNM staging (P<0.05). ③ In the breast cancer tissue, OPN, Brk, and VEGF had positive correlation with each other (P<0.05), but not in the adjacent (2cm) normal breast tissue and the distal (>5 cm) normal breast tissue (P>0.05). Conclusions The expressions of OPN and Brk from the same signal pathway increase by turns in the distal (>5 cm) normal breast tissue, adjacent (2cm) normal breast tissue, and breast cancer tissue. OPN induced the adhesion and migration of endothelial cells to accelerate vascular repair through VEGF and Brk has correlation with the progress of tumor invasion and metastasis through participating in tumor vascularization.

        • The Expression and Clinical Significance of Livin and Caspase-3 in Patients with Middle Ear Cholesteatoma

          ObjectiveTo study the expression of inhibitor of apoptosis proteins (Livin) and aspartate-specific cysteine protease-3 (Caspase-3) in patients with middle ear cholesteatoma and its clinical significance. MethodWe selected 51 patients with cholesteatoma of the middle ear treated between April 2013 and March 2014 in our department to be our study subjects. Streptaridin-perosidase immunohistochemical method was adopted to detect the expression of Livin and Caspase-3 in the middle ear cholesteatoma epithelium and normal skin of external acoustic meatus. SPSS 17.0 software package was used for statistical analysis. ResultsThe expression of Livin in cholesteatoma epithelium was significantly higher than that in the normal skin tissue of the external auditory canal (P<0.05), and the expression of Caspase-3 in cholesteatoma epithelium was significantly higher than the normal skin tissue in the external auditory canal (P<0.05). The expression of Livin and Caspase-3 in cholesteatoma epithelium was positively correlated (r=0.49, P<0.05). ConclusionsThere is a balance between apoptosis and inhibition of apoptosis in normal tissues, and when there is abnormal expression of Livin and Caspase-3 in normal tissues, it will cause cell apoptosis and apoptosis-inhibitory balance disorders, which causes middle ear cholesteatoma.

        • Expression of E-selectin of adhesive molecule in retina of rats with diabetic retinopathy

          ObjectivesTo investigate the expression of E-selectin of adhesive molecule in microvessels in rats′ retina with diabetic retinopathy (DR) at the early, middle, and late phase.MethodsNinety Sprague Dawley (SD) male rats were randomly divided into normal control group (6, 9, and 12 months subgroups with 10 rats in each subgroup) and streptozotocin (STZ) DR group (6, 9, 12 months subgroups with 20 rats in each subgroup). The DR model was set up by one-off celiac injection with STZ at a dose of 60 mg/kg. The rats were executed according to the different time points, and the eyeballs were removed and the digested stretched preparations of retinal microvessels were made. E-selectin of adhesive molecule in microvessels was revealed by EnVision immunohistochemical staining, and the expression of E-selectin in retinal microvessels and the morphological changes were analyzed and observed by Leica-Q550W computer image analytical equipment. The relativity between degree of impairment of DR and expression of E-selectin was detected.ResultsMorphological changes: hyperplasia of endotheliocytes and decreased pericytes were found in 6month DR group; irregularly arranged retinal capillary networks, obviously thickened basilar membrane, many aggregation and adhesion of leucocytes to capillary walls, large areas of acellular capillaries, and opoptosis of endotheliocytes and pericytes were found in 9-and 12-month DR group. Results of quantitative analysis: there was no expression of E-selectin in normal control group, while expression of E-selectin with different degree in each DR group was found. There was significance difference among DR subgroups (Plt;0.01), and the highest expression was found in 6-and 12-month DR group.ConclusionsThe expression of adhesive molecule in microvessels in rats' retina increases as the disease course of DR extends, and has positive correlation with the degree of impairment of microvessels in DR.         • Expression of connective tissue growth factor in injured model of retinal pigment epithelial cells and the promoting effect on migration

          ObjectiveTo observe the expression of connective tissue growth factor (CTGF) in injured model of retinal pigment epithelial (RPE) cells and the promoting effect of CTGF on migration of RPE cells.MethodsCultured monolayer-confluent human RPE cells were scraped with a trephine and a cotton stick, and set up the injured model of RPE cells with round scraped area. Immunohistochemistry and in situ hybridization(ISH) were used to detect the expression of CTGF protein and mRNA in injured RPE cells at distinct time points after injury. The number of RPE cells migrated to injured area was measured and the effect of CTGF on migration of RPE cells and the effect of dexamethasone (DEX) on the promoting process of CTGF were observed.ResultsThe results of immunohstochemistry and ISH indicated the weak positive expression of CTGF in RPE cells at the edge of scrape 6 hours after injury, and the positive expression increased gradually as time goes by after the injury. Strong positive expression of CTGF in RPE cells at the edge of scrape was found 24 and 48 hours after injury. Rebuilt human CTGF stimulated migration of RPE cells in a dose-depended manner, and DEX significantly inhabited the migration.ConclusionCTGF involves in the procedure of repair of injury of RPE cells, which may play an important role in the pathogenesis of intraocular proliferative diseases such as proliferative vitreoretinaopathy.(Chin J Ocul Fundus Dis, 2005,21:306-309) 

        • The correlation among connective tissue growth factor,transforming growth factor-β receptor,and extracellular matrix in human proliferative membranes of proliferative vitreoretinopathy

          Objective To investigate the expression of connective tissue growth factor(CTGF)in human proliferative membranes of proliferative vitreoretinopathy(PVR),and the relationship among CTGF,transforming growth factor-beta; receptor(TGF-beta;R)and extracellular matrix(ECM). Methods Immunohistochemistry method of streptavidin-biotin-peroxidase complex(SABC)was used to detect the expression of CTGF,TGF-beta;RⅡ,fibronectin(FN),collagen Ⅰ,and collagen Ⅲ protein in43periretinal membranes(PRM)of PVR obtained by vitrectomy,and the correlations of the expression of CTGF,TGF-beta;RⅡ and ECM were analyzed by statistics. Results CTGF and TGF-beta;RⅡ protein highly expressed in PRM of PVR and most of the CTGF-positive cells were epithelial cells.The result of immunohistochemistry showed that the positive rates of CTGF and TGF-beta;RⅡ protein were 70.6% and 76.5%in PVR C membranes,and 73.9% and 69.6%in PVR D membranes respectively.Relationship between positive expression and membranesprime; grades appeared no statistical correlation(P>0.05).Statistical analysis showed that there was a correlation between the expression of CTGF and TGF-beta;RⅡ,FN,and collagen Ⅰ and Ⅲ protein,respectively. Conclusions The expression of CTGF and TGFbeta;RⅡ protein is up-regulated in PRM of PVR,which suggests that the activation of TGF-beta;RⅡ is involved in the production of CTGF,and CTGF is closely related to the production of ECM and play an important role in the pathogenesis of PVR. (Chin J Ocul Fundus Dis, 2006, 22: 192-195)

        • Expression and Clinical Significance of TGF-β1 and p27 in Gallbladder Carcinoma

          【Abstract】Objective To investigate the expression of tumor growth tactor β1 (TGFβ1) and p27 in gallbladder carcinoma and their relation to the development of the carcinoma. Methods The expression of TGF-β1 and p27 in 36 cases of gallbladder carcinoma was detected by SP immunohistochemical staining. Twenty cases of chronic cholecystitis were collected as control. Results The positive rate of TGF-β1 (63.9%) was higher than that of the control (10.0%),P<0.05, and the positive rate of p27 (47.2%) was lower than that of the control 100%(P<0.05). The positive rate of TGF-β1 was significantly higher in metastasis or Nevin Ⅳ~Ⅴ stage cases than that of non-metastasis or Nevin Ⅰ~Ⅲ stage cases 33.3% (P<0.05). The positive rate of p27 was statistically higher in moderate and highly differentiation (60.9%), nonmetastasis (75.0%) or Nevin’s Ⅰ~Ⅲ stage (75.0%) cases than those of poor differentiation (23.0%), metastasis (33.3%) and Nevin Ⅳ~Ⅴ stage (33.3%) cases (P<0.05). The expression of p27 was negatively correlated with that of TGF-β1(r=-0.4473,P<0.05). There was significant difference in survival time between patients with TGF-β1 positive and TGF-β1 negative(P<0.05). The difference was also found between patients with p27 positive and p27 negative. Conclusion The upregulation of TGF-β1 and downregulation of p27 in gallbladder carcinoma indicates the imbalance of TGF-β1/p27 system, which may play a role in the carcinogenesis and predict the malignant behaviors of the carcinoma.

        • Expression of vascular endothelial growth factor and basic fibroblast growth factor in diabetic rats

          ObjectiveTo investigate the expression and relation of vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF) in rats with diabetic retinopathy.MethodFifty-five Wistar rats were randomly divided into the control group (10 rats), and 1, 3, and 5-month-diabetes group (15 rats in each diabetes group), and the diabetic models were set up. The expressions of VEGF and bFGF were detected by situ hybridation and immunohistochemistry on retinal paraffin sections.ResultsThe results of situ hybridation showed that expression of bFGF was found in 3-month-deatbtes group with the percentage of 77.8%, and 88.9% in 5-month-deatbtes group; the positive expression of VEGF was not found in 3-month-deatbtes group but in 5-month-deatbtes group with the percentage of 66.7%. Immunohistochemistry indicated that the positive expression of bFGF started in 3-month-deatbtes group with the percentage of 55.6%, and 88.9% in 5-month-deatbtes group; the percentage of the expression of VEGF was 33.3% in 3-month-deatbtes group and 88.9% in 5-month-deatbtes group.ConclusionThe expression of VEGF occurs after the expression of bFGF in rats with DR.(Chin J Ocul Fundus Dis, 2005,21:37-40)

        • THE EXPRESSION OF CELL CYCLE-REGULATED PROTEINS IN PANCREATIC ADENOCARCINOMA AND ITS SIGNIFICANCE

          Objective To know the abnormal expression of the cell cycle-regulated proteins in pancreatic adenocarcinoma and their effect on tumor cell growth. Methods The expression of p16, p21, Rb and p53 protein in 47 cases were investigated by immunohistochemistry with wet autoclave pretreatment for antigen retriaval. Furthermore, tumor growth index were assessed by a novel anti-ki-67 antibody (ki-s5). Results All the expression of p53, p16, p21 and Rb protein were the nuclear stainning. The positive rates of p53, p16, p21 and Rb protein were 55%, 53%, 74% and 98% respectively. There was negative correlation between of p16, p21 or Rb protein expression and ki-67 growth index. No relation of p53 protein stainning and the expression of p21 protein was found. Conclusion In pancreatic adenocarcinoma, the negative expression of p16 protein and p21 protein may play an important role in tumor cell growth, but tumor proliferation caused by abnormality of Rb protein is rare. The expression of p21 protein was not associated with the expression of p53 protein.

        • The Correlation Between Vascular Endothelial Growth Factor, Microvessel Density and the Progression of Gastric Carcinoma

          Objective To investigate the relationship of vascular endothelial growth factor (VEGF), microvessel density (MVD) and progression of gastric carcinoma (GC). Methods The expression of VEGF and MVD in archival waxembedded specimens of 80 cases of GC and 20 gastric benign disease (GBD) were examined by using immunohistochemical staining. ResultsThe positive expression rate (PER) of VEGF in GC was 75.0%, and in GBD 5.0% (P<0.05). The PER of VEGF in GC with invasive serosa was 95.5%, in those without serosal invasion 50.0% (P<0.05). 82.8% was the PER of VEGF in GC with lymph node metastasis, 54.5% without lymph node metastasis (P<0.05).The PER of VEGF in GC accompanied by distant metastasis was 100%, higher than that without distant metastasis (71.0%, P<0.05). PER of VEGF in pTNM Ⅰ+Ⅱ was 53.1%, in Ⅲ+Ⅳ 89.6% (P<0.05). MVD correlated significantly with depth of invasion, lymph node metastasis,distant metastasis and pTNM stages (P<0.05). There was correlationship between MVD and VEGF (P<0.05).Conclusion VEGF expression upregulation and MVD contribute to the progression of gastric carcinoma.

        • Study on cellular phenotype and apoptosis involved in experimental autoimmune uveoretinitis

          Objective To investigate the cellular phenotype involved in experimental autoimmune uveoretinitis (EAU) and apoptosis of infiltrating cells in this inflammation. Methods Immunohistochemical staining and in situ apoptosis staining were performed using monoclonal antibodies to monocytes and macrophages (EDI),MHC calss -II antigen (OX6),T lymphocytes (R73) and TACS 1 Klenow kit on both ocular sections and wholemounts of 16 Lewis rats after immunization with interphotoreceptor retinod-binding protein(IRBP). Results EAU was induced in 12 of 16 Lewis rats with a clinical inflammation score being 1.29plusmn;0 .7.Influx of monocytes,lymphocytes and MHC class II+ cells into the uvea and retina was noted after immunization with IRBP.Apoptosis of infiltrating cells was observed in the uvea and retina and more apoptotic cells were present in the iris and ciliary body compared with the choroid and retina. Conclusion A number of cells including monocytes,macrophages,lymphocytes and MHC class II+ cells are involved in EAU induced by IRBP.Apoptosis of infiltrating cells occurs at early stage of EAU,which may greatly contribute to the rapid regression of the inflammation induced by IRBP.
