        • Association among Fructosamine and Blood Glucose, Glycated Hemoglobin

          【摘要】 目的 探討果糖胺(fructosamine,FMN)與血糖、糖化血紅蛋白(HbA1c)的關系。 方法 2009年5月-2009年8月,以75例糖尿病患者作為糖尿病組,36例健康志愿者作為正常對照組,進行口服糖耐量試驗,檢測其空腹血糖及餐后2 h血糖水平,并同時檢測FMN與HbA1c水平。 結果 糖尿病組空腹血糖、餐后2 h血糖、FMN、HbA1c均高于正常對照組(Plt;0.05)。FMN與HbA1c、空腹血糖、餐后2 h血糖均呈正相關關系(Plt;0.05)。FMN與空腹血糖的相關系數高于HbA1c與空腹血糖的相關系數,FMN與餐后血糖的相關系數也高于HbA1c與餐后血糖的相關系數。 結論 果糖胺與HbA1c相比有一定的優勢,可作為監測糖尿病患者血糖控制的良好指標。【Abstract】Objective To investigate the association among fructosamine (FMN) and blood glucose, glycated hemoglobin (HbA1c). Methods From May 2009 to April 2009, 75 diabetes and 36 health subjects were recruited for this study. Blood samples was collected and assayed for FMN, HbA1C, and fasting glucose.The 2-hour oral glucose tolerance test (OGTT-2h) was given. Results Fasting glucose, OGTT-2h glucose, FMN and HbA1c were all higher in diabetes group than in healthy control group (Plt;0.05) .There was positive correlation between FMN and fasting glucose, OGTT-2h glucose, and HbA1c (Plt;0.05). The correlation coefficients between FMN and glucose were higher than those between HbA1c and glucose. Conclusion Compared with HbA1c, FMN has advantage in monitoring blood glucose in diabetes and be regarded as a suitable index for blood glucose control.

        • Clinical Study of Microsurgical Treatment on Patients with Anterior Communicating Artery

          【摘要】 目的 探討經翼點入路顯微外科手術治療前交通動脈瘤的方法。 方法 回顧性分析2006年1月-2007年1月間38例前交通動脈瘤患者的臨床表現,影像資料、手術治療及預后情況。所有患者均施行經翼點入路手術,夾閉38例。 結果 治愈37例,死亡1例。37例隨訪12個月均無前交通動脈瘤再出血發生。 結論 經翼點入路手術治療前交通動脈瘤可收到滿意的手術治療效果。【Abstract】 Objective To investigate the methods of microsurgical treatment with transpterional approach on anterior communicating artery. Methods The clinical manifestation,imaging data,treatment and prognosis of 38 patients with anterior communicating artery from January 2006 to January 2007 were retrospectively analyzed. All the patients had undergone microsurgical treatment with transpterional approach and closure. Results Thirty-seven patients were cured, one patient was dead. There was no one of anterior communicating artery rebleeding in 12 months follow-up. Conclusion Microsurgical treatment with transpterional approach on anterior communicating artery patients can get satisfactory surgical outcomes.

        • Expression and significance of hypoxia-inducible factor 1α in endplate chondrocytes of rats

          Objective To explore the expression and significance of hypoxia-inducible factor 1α (HIF-1α) in endplate chondrocytes, and to study the relations between HIF-1α expression and endplate chondrocytes apoptosis. Methods Eight Sprague Dawley rats were selected to obtain the L1-5 intervertebral disc endplate; the endplate chondrocytes were isolated by enzyme digestion method, and the endplate chondrocytes at passage 3 were cultured under 20% O2 condition (group A), and under 0.5% O2 condition (group B). Cell morphology was observed by inverted phase contrast microscope and cell apoptosis was detected using flow cytometry after cultured for 24 hours; the mRNA expression of HIF-1α was detected by real-time fluorescent quantitative PCR, the protein expressions of HIF-1α, Bax, and Bcl-2 by Western blot. Gene clone technology to design and synthesize two siRNAs based on the sequence of HIF-1α mRNA. HIF-1α specific RNAi sequence compound was constructed and transfected into cells. The transfected endplate chondrocytes at passage 3 were cultured under 0.5% O2 condition in group C and group D (HIF-1α gene was silenced). After cultured for 24 hours, cells were observed via immunofluorescence staining of HIF-1α, and cell apoptosis was detected using flow cytometry. Meanwhile, the mRNA expressions of HIF-1α, collagen type II (COL II), Aggrecan, and SOX9 were detected by real-time fluorescent quantitative PCR, and the protein expressions of HIF-1α, Bax, and Bcl-2 by Western blot. Results At 24 hours after culture, small amount of vacuoles necrotic cells could be observed in group A and group B; there was no significant difference in apoptosis rate between groups A and B (t=1.026,P=0.471), and HIF-1α mRNA and protein expressions in group B were significantly higher than those in group A (t=22.672,P=0.015;t=18.396,P=0.013), but, there was no significant difference in protein expressions of Bax and Bcl-2 between groups A and B (t=0.594,P=0.781;t=1.251,P=0.342). The number of vacuolar necrosis cells in group D was significantly higher than that in group C, and HIF-1α positive cells were observed in group D. The apoptosis rate of group D was significantly higher than that of group C (t=27.143,P=0.002). The mRNA expressions of HIF-1α, COL II, Aggrecan, and SOX9 in group D were significantly lower than those in group C (t=21.097,P=0.015;t=34.829,P=0.002;t=18.673,P=0.022;t=31.949,P=0.007). The protein expressions of HIF-1α and Bcl-2 in group D were significantly lower than those in group C (t=37.648,P=0.006;t=16.729,P=0.036), but the protein expression of Bax in group D was significantly higher than that in group C (t=25.583,P=0.011). Conclusion HIF-1α mRNA expression is up-regulated under hypoxia condition, which will increase the hypoxia tolerance of endplate chondrocytes. Cell apoptosis is suppressed by the activation of HIF-1α in endplate chondrocytes under hypoxia condition.

        • Expression of c-met mRNA in The Drainage from Axillary Fossa after Operation for Breast Cancer

          Objective To study the significance of c-met mRNA in axillary drainage after operations for breast cancer. Methods RT-PCR assay was used to examine c-met mRNA in axillary drainage after operations in 52 cases of breast cancer. The relationships between the expression of c-met and the tumor size, metastatic lymph nodes, the expressions of estrogen receptor (ER), progesterone receptor (PR) and c-erbB-2 were analyzed, respectively. In addition, the effect of douching operative field with 5-FU and distilled water on the expression of c-met mRNA was also analyzed. Results ①The proto-oncogene c-met mRNA could be detected in axillary drainage after operations for breast cancer by RT-PCR, and its positive rate was higher than that in routine pathological detection for micrometastasis in the axillary lymph nodes (P<0.05). ②The expression of c-met mRNA was correlated with both the metastatic lymph nodes and tumor size. ③There was no significant relationship between the expression of c-met mRNA and the expressions of ER, PR and c-erbB-2. ④Dounching operative field with 5-FU and distilled water could decrease the expression of c-met mRNA.Conclusion The proto-oncogene c-met mRNA may be an ideal and specific marker for dectecting micrometastasis of breast cancer. In addition, it also suggests that the examination of c-met mRNA in the axillary drainage by RT-PCR assay could detect the micrometastasis in axillary lymph nodes much easier and more accurately than routine pathological method.
