目的 探討FasL在重癥急性胰腺炎(SAP)大鼠肺泡巨噬細胞(AM)凋亡機理中的作用。 方法 SD大鼠按數字表法隨機分為對照組、SAP組及氯化釓(GdCl3)組3組,每組16只。SAP模型制成6 h后,經支氣管肺泡灌洗獲取AM。取右肺下葉行HE染色檢查,透射電鏡觀察和雙染色法檢測AM凋亡情況,用蛋白免疫印跡法(Western blot法)檢測各組AM中FasL蛋白表達水平。 結果 GdCl3組電鏡下可見AM典型凋亡形態學特征,AM凋亡率為(22.48±1.44)%,明顯高于對照組〔(11.28±1.01)%〕及SAP組〔(6.86±1.35)%〕,其差異有統計學意義(P<0.05);GdCl3組FasL蛋白相對表達量為(1.230±0.041)%,較對照組〔(0.936±0.024)%〕和SAP組〔(0.704±0.011)%〕明顯增高(P<0.05)。AM凋亡率與AM中FasL蛋白表達水平呈線性正相關關系(R2=0.766,P<0.01)。 結論 GdCl3可能通過激活FasL蛋白表達誘導SAP大鼠AM發生凋亡。
