Abstract In order to study the influence of chitosan on bone infection, 27 New-Zealand rabbits were randomly divided into 3 groups. According to the Norden technique, the model of osteomyelitis of the tuberosity of tibia was produced experimentally by injection of staphylococcus aureus.Immediately following injection of the staphylococci, the chitosan gel, acetic acid solution and sterile distilled water were injected into the bones in the 3 groups respectively. The latter two groups were served as control. The severity of the infection was evaluated by clinical symptoms and signs, radiographicdata as well as the bone culture and bacterial counts. Compared the results from chitosan with the other two controls, it was found that the local injection ofchitosan could not reduce the incidence of bone infection, however, it could provide actual improvement when other data were concerned. The effects of chitosangel might be attributed to its bacteriostatic and immunological activity as well as its slow degradation in the body.
Motor endplate is the structure connected between the nerve terminal and muscle fibre and plays a very important role in conducting nerve impulses to the target, therefore, systemical study of the sequential changes of the motor end-plate upon denervation is quite important.Ninety New Zealand rabbits were divided into nine groups from two weeks to nine months after denervation. Acetycholinesterase(AchE) was analyzed quantitatively to study the sequential changes of the motorendplates of tibialis anterior muscle. The results showed that there was no significant reduction of AchE at theend if one month after denervation, whereas a sharp reduction of AchE afterwards. AchE could not be stained after five month denervation.
ObjectiveTo observe the dynamic changes of neuroglobin (NGB) expression in hippocampus after status epilepticus(SE) in rats, and to explore the role of NGB in epileptic seizures.Methods40 healthy male Sprague Dawley rats were randomly divided into two group according to random number table method:control group (n=5) and epilepsy model group(n=35).Epilepsy model group according to observation time was divided into:0h, 1h, 3h, 12h, 24h, 10d and 30d.Intraperitoneal injection Lithium-pilocarpine (20 mg/kg~127 mg/kg, Li-PC) to establish the rat model of SE.Observe the behavioral changes in rats with epilepsy.Nissl staining was used to detect the neuronal damage in hippocampus. Streptavidin-biotin-peroxidase complex immunohistochemical method was used to detect the expression level of NGB in hippocampus;ResultsAfter SE, the neurons in hippocampus were severely damaged with the progress of epileptic seizures, the number of surviving neurons in CA1, CA3 regions showed a near linear decline.Among them, the number of surviving neurons in (12h, 24h, 10d, 30d)CA1, (0h, 12h, 24h, 10d, 30d)CA3 and(12h, 24h, 10d, 30d) DG area were significantly lower than that of the control group (P < 0.05).The expression level of NGB in CA1, CA3 and DG region of hippocampus were increased after SE, and both of CA1 and DG were reached peak in 24h after SE, but was still higher than the control group.And the CA3 area showed a continue rising trend.Among them, CA1(24h, 10d, 30d), CA3(24h, 10d, 30d) and DG(12h, 24h, 10d, 30d) were higher than that of control group significantly (P < 0.05).In addition, it was found that there was a positive correlation between the number of surviving neurons in CA3 area and the expression level of NGB (R=0.306, P=0.011).ConclusionUp-regulation of NGB expression in hippocampus after status epilepticus, and was positively correlated with the number of neurons in the CA3 area, suggesting that up regulation of NGB expression may be a compensatory protective mechanism of ischemic injury induced by seizures, and participate in the protection of epilepsy related neuronal damage.
Application of the island flap on the back of rabbit as a model, the central vessel and its anterior edge vein was perserved. We explored the features of the blood supply and the difference in the dependence of the recipient bed of pure venous flap, arteriolised venous flap and conventional flap. The result showed that the conventional flap and arteriloized venous flap could survive, but the pure venous flap could not. It was suggested that the pure venous flap was in an impending necrotic condition, therefore,the blood circulation of recipient bed and the rate of revascularization between the recipient bed and the flap seemed to play an important role in the survival of the flap.

Hyaluronic acid (HA) is an extracellular matrix having a high molecular weight. This study is to investigate (1) whether the ultrapure HA had effect on the healing of full thickness skin wounds in animals, and (2) the relation between the effect of HA and the size of the molecular weight of HA. The ultrapure HA with different molecular weights and HA with intermediate molecular weight were applied topically to the wounds from removal of full thickness of skin and topical application of normal saline was used as control. At 21st, day after the topical application, the tensile strength was reduced in wounds of groups of HA with high and intermediate molecular weights, but not in the group of low molecular weight. Using laser-Doppler scanning of the dynamic changes of the wound perfusion, in the 3rd day the blood supply to the wound was reduced in HA groups of high and intermediate molecular weights, but at the 7th day, the blood supply of all wounds in the two groups was increased. The study revealed that HA with different molecular weights had different actions on the contraction of wounds, the angiogenesis of the granulation tissues and the healing strength of the wounds. It was indicated that hyaluronan could be used for wound treatment or as a carrier, but its accurate composition must be carefully noted.
In order to investigate the function of epidermal growth factor (EGF), the following experiments were performed. Thirty white rats were chosen and divided into 3 groups. In the back of each rat, two 2 cm x 2 cm wounds were made bilaterally, the skin and subcutaneous tissue was removed. EGF were used in one of the two wounds randomly, while those without EGF usage in control. After 1, 2 and 3 weeks, the rats were sacrificed. The area of the wounds was measured, and the healing time of each wound was recorded. The results showed that the healing time of EGF group was 14.6 days while that of control group was 18.5 days (P lt; 0.01). Furthermore, the DNA, protein and hydroxyproline contents of EGF group were higher than those of the control group (P lt; 0.01). It was suggested the EGF could accelerate wound healing and shorten the healing time.
To observe the changes of intestinal bacteriology in acute necrotizing pancreatitis (ANP). Dog ANP model was induced by injection of sodium taurocholate with trypin into the pancreatic duct. All dogs were sacrificed on the seventh postoperative day, mucosal and luminal microflora of intestine were analyzed quantitatively. The blood and organs were collected for culture. The results showed that population levels of E.coli in the intestinal mucosa and the content in cecum of the ANP dogs showed much higher level than those of the controls (P<0.01 or P<0.05), while bifidobacterium and lactobacillus were decreased significantly (P<0.01), resulting in reversal of bifidobacterium/E.coli ratio. Blood levels of endotoxin were 1-2 times higher in ANP group as compare with the controls. The positive rate of blood and organs were 100% in ANP dogs. E.coli were the major bacteria cultured. The results indicated that microecological disturbance could take place after the onset of ANP, which may take an important role on pancreatic infection complicating ANP.
Local hypothermia as a preventive method to reperfusion injury of skeletal muscles was studied. Sixteen Japanese rabbits were divided into four groups at random. Before the tourniquet was inflated, a cold gel pack was applied to the right hind leg of each rabbit for 15 minutes to produce local hypothermic condition, without application of tourniquet the left hind limb was under local hypothermic condition as a control. The duration of tourniquet ischemia was 4 hours, and then reperfusion for one and two hours in the A and B groups respectively; in the C and D groups the duration of ischemia was 5 hours, and reperfusion for one and two hours, respectively. The muscle temperature averaged 16.6 degrees C with a needle thermocouple in the hind limb under local hypothermia. The serum K+, LA, SOD, LPO were determined from bilateral femoral veins, and electron and light microscopic studies of sural muscles were done in the post-reperfusion period. It was found that the K+, LA, LPO were lower than that of the control groups (P lt; 0.01), but SOD was higher than that of the control group (P lt; 0.01). Electron and light microscopic studies showed sight but reversible damage of muscular structure with the possibility of in the hypothermic groups cell regeneration. Basing on this experimental results, this method was applied in 45 cases reparative and reconstructive surgery of limbs. The duration of application of tourniquet averaged 2 hours and 57 minutes, the longest being 4 hours and 31 minutes, when the muscle temperature had reduced to 22.4 degrees C. There were no postoperative complications associated with this technique. Local hypothermia appeared to be a safe and effective method of decreasing the reperfusion damage after ischemia.
Objective To explore the effects of mechanical stretch with variant frequencies on the alignment and differentiation of the multilayer myotubes cultured in vitro, and to select the optimized cultural condition of regenerative skeletal muscle tissue with stress loading cultured in vitro. Methods C2C12 myoblasts cultured in vitro in the groove casts of Sylgard 184 were induced into the multilayer myotubes. Meanwhile the myoblasts were treated with various mechanical stretch withcells tensile instrument, at the amplitude of 10% and the frequency of 0 (group A), 0.25 (group B), 0.50 (group C), and 1.00 Hz (group D) for 1 hour, 3 times a day. The myotubes morphology was observed by inverted phase contrast microscope at 5, 7, and 10 days after continuous mechanical stretch. And the expressions of mRNA for myogenic differentiation antigen (MyoD), Myogenin, Desmin, and myosin heavy chain (MyHC) were detected by RT-PCR and real-time fluorescent quantitative PCR (QRT-PCR), respectively. Results The mechanical stretch could promote the al igned fusion and increase the number of myotubes. Indeed the multilayer myotubes arranged more closely in group B at 7 days. At the same group, as the time went on, the mRNA expressions of MyoD gradually decl ined in each group. There were significant differences in mRNA expressions of MyoD between 5 days and 7, 10 days (P lt; 0.05). The mRNA expressions of Myogenin, Desmin, and MyHC were highest at 7 days. There were significant differences between different time points (P lt; 0.05), except the mRNA expression of Desmin of group B between 7 and 10 days (P gt; 0.05). At the same time, with the increase of frequency, the highest mRNA expressions of MyoD, Myogenin, Desmin, and MyHC were in group B. There were significant differences at the same time between group B and the other groups (P lt; 0.05), except mRNA expression of Desmin at 5 days between groups B and C, and mRNA expression of MyHC at 10 days between groups A and B (P gt; 0.05). Conclusion Low frequency (0.25 Hz) and suitable time (7 days) periodic mechanical stretch is beneficial to the differentiation of the multilayer myotubes cultured in the groove casts of Sylgard 184, but as the stretch time goes on the aging of myotubes will be accelerated.
