目的研究選擇性環氧合酶2(cyclooxygenase-2,COX-2) 抑制劑尼美舒利對人膽管癌細胞系QBC939細胞增殖的抑制作用。
方法應用MTT比色法觀察尼美舒利對QBC939細胞增殖的影響，細胞凋亡采用透射電鏡及流式細胞儀檢測，應用免疫組化法檢測尼美舒利對QBC939細胞COX-2及增殖細胞核抗原（PCNA）蛋白表達的影響。
結果尼美舒利呈時間、劑量依賴性抑制QBC939細胞的增殖，降低QBC939細胞COX-2和PCNA的表達,流式細胞儀檢測結果顯示，隨藥物濃度增加，細胞凋亡率顯著增加，透射電鏡下可見典型的細胞凋亡形態學改變。
結論尼美舒利顯著抑制QBC939細胞的增殖，誘導其凋亡，且呈時間、劑量依賴性，其機理可能與其下調細胞內COX-2蛋白表達有關。
