目的研究survivin反義寡核苷酸(ASODN)對膽囊癌細胞凋亡的誘導作用。
方法采用脂質體介導survivin ASODN轉染人膽囊癌細胞株GBC-SD，采用流式細胞儀技術檢查細胞凋亡變化，采用電鏡技術觀察細胞的形態變化，采用逆轉錄-聚合酶鏈反應（RT-PCR）檢測survivin基因表達的變化。
結果脂質體介導轉染survivin ASODN后，膽囊癌細胞內survivin mRNA表達相對系數為0.512±0.011,明顯低于未轉染者的0.980±0.012 (P＜0.01)，平均下調了47.83%(P＜0.01)，凋亡細胞比率為(26.28±3.91)%，明顯高于未轉染者的（0.50±0.23）%，P＜0.01，電鏡下可見膽囊癌細胞呈典型凋亡樣改變。
結論survivin ASODN轉染后能下調survivin的表達，可有效誘導GBC-SD細胞凋亡。
