目的探討5-氟尿嘧啶（5-fluorouracil,5-FU）體外誘導直腸癌HR8348細胞凋亡的作用及細胞凋亡與bcl-2、bcl-xl、bax及p53表達的關系。方法經5-FU處理HR8348細胞24 h后，用甲基綠-派若寧染色法和TUNEL法檢測細胞凋亡情況，并用SP免疫組織化學法檢測HR8348細胞中bcl-2、bcl-xl、bax和p53的表達。結果 HR8348細胞經5-FU作用24 h后，細胞凋亡指數（AI）明顯高于對照組，差異有顯著性意義（P＜0.01）。5-FU作用不同時相bcl-2的表達評分結果與對照組比較差異均無顯著性意義；不同時相bcl-xl的表達評分結果與對照組比較稍降低；而bax的表達評分結果隨時間延長明顯增加，24 h、36 h的表達評分結果明顯高于對照組（P＜0.01），而p53在5-FU組和對照組各時相均未見表達。結論 5-FU具有誘導HR8348細胞凋亡的作用； 5-FU可能通過上調bax的表達并改變bax/bcl-xl的比值而誘導HR8348細胞凋亡。
