目的研究組織因子途徑抑制物-2(TFPI-2)基因對人胰腺癌細胞系Panc-1細胞侵襲能力的影響.方法通過脂質體法將TFPI-2基因轉染到Panc-1細胞, 用RT-PCR和Western blot分別檢測轉染前、后TFPI-2 mRNA和相應蛋白的表達.Boyden小室法測定Panc-1-TFPI-2、Panc-1-V和Panc-1-P 3組細胞的體外侵襲能力的變化,同時將3組細胞分別接種裸鼠,觀察其體內腫瘤生長及轉移情況.結果轉染成功的Panc-1細胞有TFPI-2 mRNA及相應蛋白的表達; Panc-1-TFPI-2組、Panc-1-V組和Panc-1-P組細胞穿膜細胞數分別為(24.4±3.5)個、(61.3±4.1)個和(60.2±3.9)個,前者明顯少于后兩者(P＜0.05),提示Panc-1-TFPI-2組細胞的體外侵襲能力下降.3組細胞分別接種裸鼠,Panc-1-TFPI-2組腫瘤體積為(438.0±69.8) mm3,與Panc-1-V組的(852.0±102.9) mm3和Panc-1-P組的(831.0±78.1) mm3相比明顯縮小,差異有統計學意義(P＜0.05); 鏡下見Panc-1-V組和Panc-1-P組腫瘤組織浸潤到肌層,有肝、肺轉移灶,而Panc-1-TFPI-2組未見肌層浸潤及肝、肺轉移.結論 TFPI-2基因表達可抑制胰腺癌細胞系Panc-1的侵襲轉移能力,為胰腺癌的基因治療提供了實驗依據.
