目的　觀察T 淋巴細胞瘤侵襲轉移誘導因子1 ( Tiam 1) 反義寡核苷酸(ASODN) 轉染對胃癌細胞形體重塑的影響。方法　采用層粘連蛋白黏附法,由胃癌MKN245 細胞株(M0 ) 中篩選獲得高侵襲轉移亞株(MH ) 。以脂質體介導將Tiam 1 ASODN 轉染至MH 細胞中,并應用逆轉錄聚合酶鏈反應(RT2PCR) 及流式細胞技術分別檢測Tiam 1 mRNA 和蛋白的表達。采用HE 染色、細胞骨架蛋白染色及掃描電鏡技術觀察轉染與未轉染MH 細胞在形態學方面的不同。結果　與未轉染組相比,應用0. 43μmol/ L Tiam 1 ASODN 轉染可特異性抑制胃癌MH細胞中Tiam 1 mRNA 和蛋白的表達( P lt; 0. 01) ; 轉染MH 細胞的膜表面突起及偽足變稀疏或縮短,細胞骨架結構紊亂程度降低,斑點狀肌動蛋白小體減少。結論　特異性ASODN 轉染可有效抑制Tiam 1 在胃癌細胞中的表達及胃癌細胞形體重塑,這可能對胃癌細胞的侵襲轉移產生影響。
