目的:研究人參皂甙Rg 1對原代培養胎鼠腦神經細胞存活和可塑性的影響。方法:實驗分為:實驗組(人參皂甙Rg 1 1 mg/L,10 mg/L,100 mg/L),陽性藥物對照組(bFGF 20 μg/L)以及空白對照組。相差倒置顯微鏡觀察細胞生長情況,并測量細胞突起的長度;用MTT法測定培養細胞的存活率;Western-blot法檢測神經生長相關蛋白GAP-43和神經絲蛋白NF-200的表達。結果:(1)細胞平均突起長度:實驗高中劑量組神經元突起的平均長度均長于對照組。(2)MTT值:實驗高中低劑量組的灰度均明顯大于對照組。(3)GAP43和NF 200的表達:實驗高中劑量組的蛋白表達均明顯大于對照組。結論:人參皂甙Rg 1對于體外培養的胎鼠腦神經細胞的存活有較強的維持作用,并能促進突起生長,使神經可塑性相關蛋白表達上調。
