目的：研究轉化生長因子β1（TGF-β1）促進Tenon囊成纖維細胞的增殖作用及核心蛋白多糖（decorin）對TGF-β1促Tenon囊成纖維細胞增殖的影響。方法：（1）取青光眼患者的Tenon囊組織，采用組織塊貼壁培養法進行原代培養細胞培養，細胞經過冷凍與復蘇后，觀察細胞形態結構與生物學特性。（2）用不同濃度(0.01、0.1、1、5、10ng/mL)TGF-β1作用于Tenon囊成纖維細胞48 h。（3）終濃度為5 ng/mL的TGF-β1 和不同濃度(0.01、0.1、1、2、5μg/mL)的decorin共同作用于Tenon囊成纖維細胞48h。MTT比色法檢測Tenon囊成纖維細胞增殖。結果：（1）體外成功培養Tenon囊成纖維細胞，細胞生長活躍，形狀穩定，細胞經過冷凍與復蘇后，仍能保持其原來的細胞形態結構與穩定的生物學特性。（2）1～10ng/mL TGF-β1能促進Tenon囊成纖維細胞的增殖，并與濃度成正比。（3）2～5μg/mL decorin 能抑制TGF-β1促Tenon囊成纖維細胞增殖作用，其抑制作用與濃度成正比。結論： TGF-β1可刺激Tenon囊成纖維細胞增殖，Decorin能抑制TGF-β1促進細胞增殖的作用，其機理可能通過與TGF-β1結合抑制青光眼濾過手術后瘢痕形成。
